Fe protein over-expression can enhance the nitrogenase activity of Azotobacter vinelandii

submitted by chonkezano 1 year and 1 month ago
The effects of over-expression of NifH (Fe protein) on nitrogenase activity in Azotobacter vinelandii UW cells were studied by expressing an extra nifH gene under the control of the inducible meta -toluic acid pathway promoter Pm. The total amount of protein in UW/pJB654-N reacting to anti-NifH antibody was 2–3 fold of that in control UW when both the strains were grown to exponential phase in the presence of 4 μM m -toluic acid. As a consequence UW/pJB654-N showed two-fold higher acetylene reduction activity and released 7% higher amounts of ammonium into the growth medium than the control. Concomitant changes were observed also in the cellular levels of siderophores and iron superoxide dismutase (FeSOD). Thus, our results indicating that increased level of Fe protein in the cell can enhance nitrogen fixation activity of A. vinelandii may have biotechnological significance.

The role of the GAF and central domains of the transcriptional activator VnfA in Azotobacter vinelandii
submitted by sagota 1 year and 10 months ago - Topic: Biology
VnfA is a transcriptional activator required for the expression of the structural genes encoding nitrogenase-2 in Azotobacter vinelandii. VnfA consists of three domains: an N-terminal regulatory domain termed GAF, including a Cys-rich motif, a central domain from the AAA+ family, and a C-terminal...

The cytoplasmic cyclophilin from Azotobacter vinelandii interacts with phosphate acetyltransferase isoforms enhancing their in vitro activity
submitted by j5onjin 1 year and 9 months ago - Topic: Biology
Cyclophilins belong to the peptidyl-prolyl cis/trans isomerase family of enzymes (EC 5.2.1.8), which accelerate protein folding by catalysing the cis/trans isomerisation of proline imidic peptide bonds. In the present study, by a combination of bioinformatics methods, we identify phosphate acetyl...

How does hypoxia inducible factor-1α participate in enhancing the glycolysis activity in cervical cancer?
submitted by somonih 3 months ago - Topic: Health
The objective of this study is to explore the role of hypoxia inducible factor-1 (HIF-1) in glycolysis activity and its relationship with malignant biologic behaviors of cervical cancer. Immunohistochemistry was performed to study the protein expression and distribution of HIF-1α and glucose tra...

PCSK9 plays a significant role in cholesterol homeostasis and lipid transport in intestinal epithelial cells
submitted by koloemade 3 months ago - Topic: Health
The proprotein convertase subtillisin/kexin type 9 (PCSK9) regulates cholesterol metabolism via degradation of low-density lipoprotein receptor (LDLr). Although PCSK9 is abundantly expressed in the intestine, limited data are available on its functions. The present study aims at determining wheth...

Hypoxia Modulates A431 Cellular Pathways Association to Tumor Radioresistance and Enhanced Migration Revealed by Comprehensive Proteomic and Functional Studies
submitted by zeomolek 3 months ago - Topic: Biology
Tumor hypoxia induces cancer cell angiogenesis, invasiveness, treatment resistance, and contributes to poor clinical outcome. However, the molecular mechanism by which tumor hypoxia exerts a coordinated effect on different molecular pathways to enhance tumor growth and survival and lead to poor c...

The serine/threonine kinase Akt/PKB promotes cancer cell growth and invasion through several downstream targets. Identification of novel substrates may provide new avenues for therapeutic intervention. Our study shows that Akt phosphorylates the cancer related transcription factor Runx2 resulting...

Expression of NTRK1/TrkA affects immunogenicity of neuroblastoma cells
submitted by larrindosl 3 months ago - Topic: Health
High levels of the NTRK1/TrkA receptor are expressed in low-stage neuroblastomas, which are characterized by a good patient prognosis and often undergo spontaneous regression. In addition to apoptosis, tumor immune responses might contribute to this regression. We hypothesized that TrkA expressio...

Enhancing Cytokine Induced Killer Cell Therapy of Multiple Myeloma

Cytokine induced killer (CIK) cells are in clinical testing against various tumor types including multiple myeloma. Here, we show that CIK cells have activity against subcutaneous and disseminated models of human myeloma (KAS-6/1) which can be enhanced by infecting the CIK cells with an oncolytic...

Inhibition of PI3K/AKT/mTOR Pathway Enhances Temozolomide-Induced Cytotoxicity in Pituitary Adenoma Cell Lines in Vitro and Xenografted Pituitary Adenoma in Female Nude Mice

Invasive pituitary adenomas (PAs) are often refractory to standard therapy and salvage treatment with temozolomide (TMZ). Hyperactivation of the phosphoinositide 3-kinase (PI3K)/AKT/mammalian target of rapamycin (mTOR) pathway contributes to chemotherapy resistance in many cancers. XL765, a novel...
